- Semax is a synthetic seven-amino-acid peptide made by adding a Pro-Gly-Pro tail to ACTH(4-7), a fragment of adrenocorticotropic hormone that affects attention and memory in animals without acting on the adrenal gland. It was developed at the Institute of Molecular Genetics in Moscow and is registered in Russia as a nasal solution; elsewhere it is a research compound.
- What does the research on Semax actually show?
- In rodents, increased BDNF and TrkB expression in the hippocampus, broad gene-expression changes after experimental stroke, activation of dopamine and serotonin systems, and improved learning and attention. In humans, small Russian studies report improved attention in volunteers and better outcomes in stroke and vascular cognitive impairment. There is no double-blind, placebo-controlled trial in the Western literature.
- What is the half-life of Semax?
- Short. Rodent data indicate the intact peptide is degraded in blood within minutes, although the Pro-Gly-Pro tail slows this relative to the unmodified ACTH fragment. Effects on gene expression persist for hours to days after a single dose, which is why behavioural findings outlast the peptide.
